Rim Screws

I need to install some rim screws on my rear slicks. What is the proper way to install them? like do you put the tube in and air them up, then drill the holes for the screws? or do you leave the tub out and then drill the holes for the screws? I have a template for the screw holes. Thanks

Best to drill the holes before mounting the tires so you can make sure there are no burrs to keep the bead from seating good on wheels.
